Early selection and propagation of embryogenic callus using electrophoretic characteristics of barley callus

This experiment was conducted understand the electrophoretic characteristics between embryogenic(E) and nonembryogenic(NE) calli in order to fine out the way to select E callus at the early culturing stage. For this experiment, callus was induced from immatured embryo culture of Hordeum vulgare. Little difference in electrophoretic characteristics within the same type of the callus was observed among the three genotypes. Significant differences, however, in the amounts of proteins and the protein banding patterns in calli were observed between E and NE calli. The amounts of protein per milliliter in E calli of Bruce, Klages and Rodeo were 510 ㎍, 420 ㎍ and 430 ㎍, while those of NE calli were 240 ㎍, 175 ㎍ and 190 ㎍, respectively. Difference of total protein banding pattern between E and NE calli was observed at 68, 38, 24 and 18 kDa by SDS-PAGE. Moreover, total proteins revealed the changes between E and NE calli were also detected at 68 kDa, 38-30 kDa, 24-22 kDa and 18-15 kDa, ranging from PI 3-10, on the two-dimensional electrophoresis. These results suggest that the changes in the total proteins observed in the two types of callus, E and NE, are probably related to the formation of embryogenic callus.
