Synchronization, storage and regeneration of somatic embryos in suspension culture of ligusticum chuanxiong

This study was carried out to investigate the synchronization of somatic embryogenesis for rapid mass production of somatic embryos in suspension culture of Ligusticum chuanxiong. Suspension culture was better than solid culture for high frequency of somatic embryos. The optimum combination of inoculating cells and medium volume was 3㎖ of inoculating cells in 20㎖ of medium or 6㎖, in 40㎖, respectively. When spermidine was added, somatic embryo development was increased in concentration of 30㎎/ℓ but the frequency of abnormal embryos was also increased. Adding of spermine inhibited somatic embryo formation. ABA (0.05㎎/ℓ) treatment after twenty days of culture 20㎎/ℓ inabenfide treatment after three weeks of culture was the most effective for synchronization. Uniconazol treatment had no effect on synchronization of somatic embryo formation. Inositol (1g/ℓ) as osmotic stabilizer enhanced somatic embryo formation and synchronization, however, mannitol had no effect. Somatic embryos could be stored for 2~3 months long at 4℃.
