Factors affecting the formation of embryogenic callus and somatic embryo from immature embryo of Ranunculus sceleratus are described. Immature seeds were classified into two stages such as stage Ⅰ (5 days after antheses) and stage Ⅱ (10 days after anthesis) and cultured for germination of immature embryos. Stage Ⅰ was globular embryo and that of stage Ⅱ was cotyledonary one. Immature embryo of stage Ⅰ was more effective than that of stage Ⅱ in the formation of callus and embryos. Indirect somatic embryogenesis via callus was obtained and somatic embryos formed directly on the hypocotyl or the surface of cotyledons passed through typical stages of embryogenesis. Embryogenic potential was maintained by secondary embryogenesis through the successive generations of embryos. The induction of callus was most effective on MS medium with 0.5 ㎎/L NAA and 1.0 ㎎/L BA and the formation of somatic embryo was on the medium with 1.0 ㎎/L NAA and BA respectively. Plantlets regenerated from somatic embryo had phenotypically normal shoots and roots.
