Anther culture of Bupleurm falcatum L. was carried out to obtain the basic information on development of in vitro breeding materials. The optimum stage for callus formation was the uninucleate stage and its frequeny was 11.3%. The callus formation frequency was differed from the plantlet phenotype, that of the normal type was 3.1%, the multi-branched type was 0.4% and the few branced type was 15.6%, respectively. The callus formation frequency was 22~39% in the MS media containing of 2, 4-D 2.0㎎/ℓ and kinetin 20㎎/ℓ as a growth regulator. In the case of a heat shock at 35℃ for 2 days, the callus induction rate was much higher than that of immediately cultured. The highest callus formation frequency was 16.6% in the B₅ media containing of NAA 1.0㎎/ℓ and BA 2.0㎎/ℓ of the tested 5 different media. The callus formation frequency was 18% in the media supplemented with 3% glucose as a carbon source. The anther-derived embryoids and plantlets were regenerated in the B₅ media containing of NAA 0.5㎎/ℓ and BA 0.1㎎/ℓ.
