In vitro callus formation and plant regeneration ability of raphanus sativus lines with an extra cabbage chromosome

In order to determine the role of the C genome in in vitro culture, the callus formation and regeneration ability of intergeneric crosses between Brassica oleracea and Raphanus sativus were investigated. Vegetative organs and flower organs derived from chromosome addition lines of R. sativus containing a single chromosome of B. oleracea showed variation in callus formation and regeneration ability. In particular, addition lines A, B and G showed good callus growth as well as high regeneration frequency. This suggests that the genes associated with high tissue culture ability particularly those which control callus formation and subsequent regeneration from callus under in vitro conditions is concentrated on specific chromosomes of the C genome incorporated in addition lines A, B and G.
