Immature embryos of Wasabia japonica were isolated from immature seeds of field-grown plants (var. Dalma). Its stages were classified into torpedo shape, early and late cotyledonary stage and they were cultured on modified Murashige and Skoog (MS) media without plant growth regulators. The immature embryo of early cotyledonary stage was the most effective on direct somatic embryogenesis from cotyledon tissue. In order to examine the effect of plant growth regulators on callus induction and somatic embryogenesis, the immature embryos of early cotyledonary stage were cultured on modified MS media supplemented with different concentrations of auxin alone or combinations of auxins and BA. Somatic embryos were all differentiated from cotyledon tissue of immature embryo, but hypocotyl and radicle did not form callus or somatic embryos at all. The media supplemented with 1.0 or 2.0 ㎎/L IAA was the most effective on direct somatic embryogenesis. 2, 4-D and NAA were more effective than IAA on somatic embryogenesis from callus. But the combinations of auxins and BA seemed to have nothing to do with somatic embryogenesis. The formation of multiple shoots was the best on medium with 2.0 ㎎/L kinetin plus 0.1 ㎎/L IAA and a lot of plantlets regenerated from somatic embryos had phenotypically normal leaves and roots.
