Chromosome doubling of solanum tuberosum dihaploid - S. chacoense hybrids through in vitro adventitious bud formation in potatoes

An in vitro adventitious bud technique with the use of tissue culture method was adopted for chromosome doubling of dihaploid-species hybrids(2n=2x=24) obtained from interspecific crosses between dihaploids derived from S. tuberosum cv. Atlantic and selected lines of S. chacoense. Their rachises, petioles and leaflet blades were used for in vitro adventitious shoot formation. Seventy five of 163 explants regenerated were chromosome-doubled tetraploid(2n=4x=48) plants. The regeneration rate and chromosome doubling efficiency were largely affected by the genotypes of clones and different explant types used.
