Factors affecting growth and indigo content of hairy root in tissue culture of polygonum tinctorium lour

This study was carried out to get better information about the medium and its composition for hairy root growth and indigo production. The results indicated that : SH liquid medium was better than half-strength SH and MS media. 2,500㎎/ℓ nitrate plus 150㎎/ℓ ammonium nitrogen combination was more effective than other combinations used for hair root growth and indigo formation. Sucrose was more better than glucose for indigo production and its the highest was at 3% level while no difference in growth rate between them at the same level. Optimum pH for indigo production was 5.7. Addition of 200㎎/ℓ chitosan and 10U/ℓ or 20U/ℓ pectinase increased indigo formation while better hairy root growth was observed in add of pectinase only.
