Meristem culture for healthy seedling production in dioscorea batatas

The studies were aimed to establishment of the systems for healthy seedling production and mass propagation technique through the meristem culture, and the seedling rasing methods of transplantation in Dioscorea batatas DECNE through the tissue culture. In bulbil culture, the most desireable method for sterilization through this experiment was at the double treatment in the concentration of 1% benlate-T(benomyl+thiram, commercial) and 20% Lax(4% Sodium hypochloride, commercial) solution. With the supplemented treatment at the concentration of 3㎎/ℓ of kinetin(6-furfuryl amino purine) was the most effective for sprouting and plant growth and development in MS media. At the concentration of 1/8 MS medium with the application of nitrate as a nitrogen element supplemented, the shoot formation was the most effective from auxiliary bud derived meristem culture. The suitable phytohormone combination for plant propagation in vitro was in the treatment of NAA(1-Naphthalene acetic acid) 1.0㎎/ℓ and kinetin 0.5㎎/ℓ in MS medium which showed 3.8 times greater in plant propagation per month. When transplanting the in vitro plantlets, the survival rate was the best in the green house condition and sandy soil. Plant growth was developed well according to wide space, 40×30㎝, of planing density.
