Hairy root culture condition for pigment accumulation in Raphanus sativus L. cv. Chungpihongsim

To produce anthocyanin pigment by hairy root culture of Raphanus sativus L. Chung-pihongsim, factors affecting the growth and anthocyanin synthesis in hairy root were investigated. NAA was good for hairy root growth but 2, 4-D was better for pigment synthesis than other auxins used. Hairy root growth and pigmrnt formation were not affected by addition of kinetin while pigment synthesis was tended to increase when combined with 2, 4-D. 5% sucrose was the most effective for hairy root growth but higher pigment synthesis was occured at 7% level. Hairy root growth was continued up to 340㎎/ℓ phosphate but pigment synthesis was decreased at higher concentrations than 85㎎/ℓ phosphate. Hairy root growth and anthocyanin production were favorable under 400lux fluorescent light than dark. As the time course of hairy root growth and anthocyanin production, the pigment production was maximum after two weeks though hairy root growth was the highest after three weeks culture.
