Artificial seed production from somatic embryos and its germination in Lycium chinense Mill

An artificial seeds was developed using somatic embryos derived from the leaf callus of Lycium chinenes Mill. Somatic embryos were singly alginate-encapsulated to be used as artificial seeds. When cultured on MS basal medium, 95% of the non-encapsulated and 49% of the encapsulated somatic embryos were capable of germination. Among various concentrations of sodium alginate tested, the best result for bead formation was obtained in 3% sodium alginate. Of all tested media, 52% of somatic embryos were germinated on 2 MS medium but up to 60% on 3/2 MS medium. Characoal supplemented to the sodium alginated solution promoted shooting and rooting. The highest germination(80%) was obtained when artificial seeds was encapsulated with sodium alginated containing 0.5% charcoal. Artificial seeds did not lose the capacity for germination even after storing at 4℃ for 45 days. Shoots and roots of the encapsulated somatic embryos were induced after 4 weeks on 3/2 MS medium without growth regulators. Whole plantlets were successfully transplanted to potting soil for futher growth.
