Tagging gene for blast resistance in rice using RFLP markers

We report the tagging of a blast(Pyricularia grisea) resistance gene, designated Pi-12(t), using RFLP markers in rice. This gene confers resistance to Korean isolate KI-313 of the blast pathogen. To know whether there is a relationship among the genes conditioning resistance to location-specific isolates of the blast pathogen and thereby to identify linked markers to resistance gene for KI-313 isolate, RFLP(Restriction Fragment Length Polymorphism) markers previously reported to be linked to major blast resistance genes in different rice germplasm and other markers mapped to nearby regions were surveyed for polymorphism between a resistant(Suweon 365) and a susceptible(Chucheongbyeo) cultivar. Linkage of markers showing polymorphism between parents with Pi-12(t) was verified using an F₂ population and its F₃ progenies. RFLP analysis pointed out that the gene is linked to a single-copy DNA clone RZ536 on chromosome 11 with a distance of 5.2 cM. Two additional markers on chromosome 11(CDO365 and RG303) were mapped to define the location of the gene. The allelism relationship between Pi-12(t) and the two genes, Pi-k and Pi-1(t) on chromosome 11 remains to be clarified. These linked markers could facilitate early selection of the Pi-12(t) locus in rice breeding program.
