This study was carried out to determine the optimum conditions for the embryo formation in the shed microspore culture of Paeonia lactiflora Pall. The highest frequency of microspore division(38.7%) and embryo formation(123 embryos/100 anthers) was obtained at the shed microspore density of 3.1×10⁴ cells/㎖. Cold pretreatment(5℃, 10 days) of anthers enhanced the division of shed microspores and embryo formation. Pretreatment of anthers at low temperature resulted in the formation of more embryos from microspores. The concentration of 3% sucrose gave better results than that of the sucrose with sorbitol in the division of shed microspores and embryo formation. The preculture of anthers on solid medium was not effective for the embryo formation from shed microspore culture of Paeonia lactiflora.
