This study was carried out to tag the ges gene of rice using DNA markers and to confirm the locus on the molecular map via saturated mapping of DNA markers on the region of the ges gene in order to facilitate the selection of the super-giant embryo character in segregating populations and to provide the foundation for map-based gene cloning. The RFLP, microsatellite and RAPD markers which were linked to ges gene were identified by bulked-segregant analysis. The twenty-six out of thirty-six RFLP markers of chromosome 7 showed polymorphism between Hwacheongbyeo-ges and Milyang 23. Four of 400 RAPD primers were screened via bulked-segregant analysis. However, F₂ analysis revealed that OP-31 of them was linked to ges gene. The base sequence of the primer OP-31 was 5’-GGAGCCTCAG-3’. All the markers as well as the embryo character demonstrated the expected segregation ratio of the single gene. RZ395 and CDO497flanked the ges gene at 2.4 cM and 3.4 cM, respectively, on chromosome 7. The two microsatellite loci, RM18 and RM10 were linked to ges at 7.7 cM and 9.6 cM, respectively, and the RAPD marker, OP-31, was linked to ges at 7.2 cM. The availability of DNA markers to facilitate the selection of this grain character and further utilization of the molecular map developed here for map-based cloning of ges gene were discussed.
