This study was conducted to determine the pattern of self-incompatibility through self-and cross-pollination in two varieties of herbaceous peony (Paeonia lactiflora Pallas). After self-pollination with the pollens of same variety, pollen tubes have germinated and grown into style tissue. However, the pollen tubes have arrested the growth in the mid of growing stage. This pattern was a different from that of sporophytic self-incompatibility. In the case of sporophytic self-incompatibility, self-pollinated pollens are not germinated on the style in normal, but if the pollens do, they do not penetrate into style tissue. From these phenomena, the pattern of self-incompatibility in herbaceous peony is assumed gametophytic self-incompatibility. For more precise research, protein analysis using SDS-PAGE and RNase activity stain gel electrophoresis was conducted. In SDS-PAGE analysis, two major proteins (21.5 and 28-kDa) was detected, and ribonuclease activity was tested in activity stain gel. Ribonucleatic activity is a unique characteristic of stylar proteins of gametophytic self-incompatibility plants reported previously. From these results, we concluded that P. lactiflora has a gametophytic self-incompatibility.
