절단된 nopaline synthase promoter의 외부 trigger에 의한 유도 활성

Induced activity of deleted nopaline synthase promoter by external trigger

The nopaline synthase (nos) promoter is regulated by various developmental and environmental factors. The nos upstream region essential for this regulation was studied by using internal deletion mutant lacking nos upstream element with synthetic oligonucleotides. Insertion of 20 nucleotide sequence (131-112) containing two hexamer motifs and a spacer region into -126/-112 internal deletion mutant recovered promoter activity of nonfunctional deletion mutant and responded to various triggers. Ascorbic acid induced the nos promoter activity significantly while H₂O₂ induced in weak level. Deletion of one or more nucleotide of either hexamer sequence reduced nos promoter activity. Point mutations of a spacer region between two hexamers altered the strength of expression of the nos promoter.
