This study was conducted to select the most effective condition for the commercial application of indigoid production and growth in hairy root of Polygonum tinctorium Lour. transformed by Agrobacterium rhizogenes strain 11325 and 15834 in the air-lift bioreactor. Growth and indigo of hairy roots was turn into the stationary phase after 20 days, which suggested that the subcultural period might be 30 days. Higher indigo and indirubin contents were detected 20 (400 g/g) and 30(215.6 g/g) days, respectively, after add of 200mg/1 indole as precursor at the beginning of culture in the air-lift bioreactor. However, only indigo was detected in the control. Pluronic F-68 was effective for growth and indigo production as elicitor at 0.0lg/l but over this level increased working volume which caused contamination the medium in air-lift bioreactor. Silicon-based antifoam C-emulsion treatment with 200ppm supress effectively foam overflowing.
