Flow cytometric characterization and sorting of cultured Bupleurum falcatum Microspores

Under the appropriate culture conditions microspores could be induced switch their development pathway from pollen to embryo formation. Many efforts are under way to elucidate the molecular, physiological and morphological changes associated with embryogenesis. A goal of our research program is to characterize those cells in Bupleurum falcatum microspore culture that are active. For this purpose we have used flow cytometric and epifluorescent techniques. The knowledge obtained from these types of studies will give us insight into early stage in plant development and may lead to the application of microsporederived from haploid plants for breeding in recalcitrant species. Activity of cultured microspore was different from the developmental stage that of the tetrad, uni-nucleate, bi-nucleate and matured pollen were 15.1, 36.4, 84.0 and 56.4%, respectively. Optimum culture medium was determined by the OM media as 79.4% of active microspore presence 9days after culture. Korea originated cultivar was more active than Japan’s cartivar “Misima” during the micorspore culture.
