RAPD mapping in interspecific cross of essex × PI 437654 in soybean

Seventy-nine F2:3 progenies from an intraspecific cross of G. max × G. max, cultivar ‘Essex’ and plant introduction PI 437654, formed the population for the construction of an RAPD map. Forty-one RFLP markers containing 21 polymorphic markers from probe/enzyme combinations and 20 polymorphisms from probe/enzyme non-combinations, and 164 RAPD markers were selected for mapping. One hundred thirty-seven markers, including 104 RAPD markers, 31 RFLP probes, and 2 morphological markers mapped into 27 linkage groups. The linkage map defined 1,096.3 cM of the soybean genome with an average map distance of 8.12 cM. Twenty-seven markers(13.0%) were significantly (P<0.05) deviated from the expected segregation ratio. Multiple RAPD polymorphic loci generated by the same primer were mapped to the same linkage group and/or different linkage groups. Additionally, polymorphisms generated by different primers were localized at the same position.
