Mutagenesis of arabidopsis thaliana by diepoxybutane and mutant analysis

Arabidopsis thaliana seeds were treated with a series of 18, 19, 20, 21, 22 mM diepoxybutane of bifunctional alkylating agent. The visible mutants were selected in M₂ population that was from the selfing of M₁ pools. The selected mutants were variegation type, chlorophyll deficiency, flower deformed type, long hypocotyl, male sterile type and so on. These mutants were crossed with wild type and showed the segregation ratio (wild type : mutant type) of 3 : 1. The ratio indicated that the mutant was due to single recessive mutation. RAPD analysis of the mutant of flower deformed type showed the polymorphism of amplified bands.
