Mapping of genes confering resistance to Korean isolates of rice blast fungus using DNA markers

Resistance genes for rice blast have been tagged and mapped in several laboratories. To know whether there is a relationship among genes conditioning resistance to location-specific isolates of the blast pathogen and thereby to identify linked markers to resistance gene for isolates KJ-101, KJ-201 and KJ-105 isolated in Korea, DNA markers including RFLP markers previously reported to be linked to major blast resistance genes in different rice germplasm and other markers mapped to nearby regions were surveyed for polymorphism between a resistant (Suweon 365) and a susceptible (Chucheongbyeo) parent. Linkage of markers showing polymorphism between parents with the resistance gene was verified using an F₂ and F₃ population. Linkage analysis showed that the resistance genes to isolates KJ-101, KJ-201 and KJ-105 were linked to single-copy DNA clones RG869 on chromosome 12, RG213 on 6 and RM164 (microsatellite marker) on chromosome 5, respectively. The relationship between the three known resistance genes Pi-2 (t), Pi-10 (t) and Pi-4 (t), on chromosomes 6, 5, and 12 respectively and resistance genes in Suweon 365 remains to be clarified.
