닭血漿중 vitamin A의 運搬蛋白質에 關한 硏究

Studies on the transport protein for vitamin A in chicken plasma

A chick was fed with vitamin A supplemented diets, namely, 1,000,000 I.U. per kg of feed for 27 days. Blood sample was collected by heart puncture and harvested blood plasma in an oxalated state. Blood plasma was fractionated for the separation of retinol transporting protein by gel filtration and the retinol transporting protein fraction eluted through the column was subjected to 1% SDS-polyacrylamide gel electrophoresis for the determination of molecular weight. The results obtained were summarized as follows. 1. By means of Sephadex G-200 gel filtration: chicken plasma was separated into 3 protein peaks, and the 3rd peak was retinol transporting protein fraction detected by the trifluoroacetic acid reaction. 2. The retinol transporting protein fraction isolated by Sephadex G-200 gel filtration contained 2 bands when subjected to 1% SDS-polyacrylamide gel electrophoresis. The molecular weight of the faster moving band of out of two was 68,000 and the other was 94,000. The later, whose molecular weight of 94,000, seemed to be the complex of 2 proteins, that is, the faster moving band and the retinol transporting protein, whose molecular weight was 26,000.
