Percent hemolysis of erythrocyte, plasma trypsin inhibitor capacity and glutathione peroxidase activity were measured in erythrocytes and plasma after 100 mg/kg po(LD 50) dose of paraquat to Wistar rats presupplemented for 4 weeks at dietary levels of 0,10,50,100, and 1,000ppm, and after access to diets containing various levels of α-tocopherol and 300 ppm paraquat water of libitum for 4 weeks. The results obtained were summarized as follows: 1. Presupplement of a-tocopherol at a concentration of less than about 10 mg/kg for 4 weeks significantly increased percent hemolysis of erythrocytes by dialuric acid. 2. Percent hemolysis of erythrocytes after a single toxic paraquat doses to rats presupplemented with various concentrations of α-tocopherol was not significantly altered from control values. 3. Chronic administration of 300 ppm paraquat water to rats presupplemented with α-tocopherol significantly increased percent hemolysis. Especially rats presupplemented with α-tocopherol of less concentrations than 10 mg/kg show-ed a remarkable elevation of percent hemolysis. 4. Plasma trypsin inhibitor capacity was significantly increased in almost of all groups of rats after paraquat treat-ment. But plasma trypsin inhibitor capacity after chronic doses of paraquat was significantly greater than that seen in a single toxic dose of paraquat, showing the protective effect of α-tocopherol against acute paraquat poisoning especially when α-tocopherol at a concentration of 1,000 mg/kg was supplemented to rats. 5. Plasma and erythrocyt glutathione peroxidas activities were not altered by chronic paraquat treatment.
