Thermophilic Campylobacter spp.의 분리방법에 관한 비교연구

In order to study the rapid methods in the isolation compare the isolation rates in various enrichment and selective media under different culture temperature and microaerobic growth condition. Also examined were the efficiency of darkfield microscopic examination (DFME) for presumptive diagnosis of thermophilic Campylobacter spp. from various animals feces. A total of 35 strains were isolated from 129 diarrheal feces. The isolation rates in direct cultures of thermophilic Campylobacter spp. from chicken, pig and cattle were 66.7%, 21.7% and 13.1%, respectively. Thirty-five specimens of a total 129 specimens used for direct cultures were presumptively diagnosed as containing Campylobacter spp. by DFME, 33 strains were recovered from these 35 specimens. The accuarcy of presumptive diagnostic value of DFME was reached 94.3%. In the comparison of cultural temperature, isolation rate in 42℃ incubation is more higher (72.5±6.2%) than that in 37℃ incubation (63.1±5.9%) (P<0.05). In comparison of microaerophilic condition, utilization of GasPak Ⅱ system (72.1±6.4%) appeared to be effective than CO₂ generation method (63.4±6.2%) (P<0.01). In the comparison of isolation rate of various enrichment media by 42℃ GasPak condition, Doyle, VTP-FBP broth and Campythio medium (94.3%) were highly effective for the isolation of thermo-philic Campylobacter spp.. In the comparison of isolation rate of various selective media by 42℃ GasPak condition Campythio BAP medium (87.2±9.5%) and MSKM (85.7±5.8%) were highly effective for the isolation of thermophilic Campylobacter spp.. Therefore, cultivation on Campy-BAP medium after enrichment in Campy-thio, Doyle and VTP-FBP broth was the most effective method in the isolation of thermophilic Campylobacter spp.. Also, cultivation of MSKM after enrichment in Doyle’s broth had some efficiency as Campy-BAP medium.
