Physico-chemical properties of normal and treated chicken embryo allantoic fluid (CEAF) which promotes the growth of NDV(B₁) in chicken embryo fibroblast monolayer were studied and the experimental results were summarized as following: 1. Normal values of CEAF harvested from 10 to 13-day-old embryos were: 1) Maximum absorption wave length in spectrophotometry: Three peaks of 210nm, 238nm and 293nm. 2) Total protein content: 1.24±0.2 to 4.15±0.03 mg/ml. 3) Osmolarity: 210±1.0 to 243±2.0 mOsm/kg H₂O. 4) pH: 8.2 to 8.5. 2. Stabilities of normal CEAF harvested from 10 to 13-day-old embryos after the treatments were: 1) Heating: Stable at 70℃ for 20 minutes heating. Unstable at 80℃ for 20 minutes heating. 2) Acid or alkaline treatment: Stable at pH 8.0 but not at pH 5.0 or 10.0. 3) Trypsin treatment: Stable in 0.01% 10 minutes treatment. Unstable in 0.05% 20 minutes treatment or 0.1% 10 minutes treatment. 4) Ether treatment: Unstable by 3-time-extraction. 3. SDS-PAGE patterns of normal and treated CEAF harvested from 10 to 13-day-old embryos were: 1) Normal CEAF: 4 bands (MW 17T, 33T, 68T and 84T). 2) Heated (80℃ 30 min.) CEAF: Same as the normal CEAF. 3) Acid (pH 4.0) treated CEAF: Same as the normal CEAF. 4) Ether (3-time-extraction) treated CEAF: Same as the normal CEAF. 5) Trypsin (0.1% 10 min.) treated CEAF: 2 bands (MW 17T and 48T). 4. The values of normal and treated CEAF of 14-day-old embryos, which showed no or less NDV growth promoting activity, were different from those of 10 to 13-day-old embryo CEAF. In general, 14-day-old embryo CEAF contained an increased protein contents which may interfere the virus growth promoting activity of CEAF.
