The effects of butylated hydroxytoluene(BHT) on paraquat-induced pulmonary lesions were examined in male rats. Sprague-Dawley male rats were divided into 2 control groups(C₁, C₂) and 3 treatment groups (T₁, T₂, T₃). The C₁ group and treatment groups were given a single oral dose of paraquat at 80 mg/kg body weight and the C₂ Group was given a single oral dose of physiological saline(1ml/rat). The rats in the C₂ groups were given con oils(1 ml/rat/day) for 5 days. Those rats treated with paraquat in the T₁, T₂, T₃ groups were given oral doses of BHT at 10, 20 and 30mg/kg body weight respectively for 5 days. The results were summarized as follows: 1. The increasing rate of body weight in C₁and T₃ groups was decreased significantly to compare with that of C₂ group(p<0.01, p<0.05, respectively). 2. The histopathological changes observed in the paraquat-treated rats were pulmonary hemorrhage, perivascular edema, focal fibrosis, infiltration of fibroblasts, desquamation of bronchiolar epithelial cells, hyperplasia and adenomatoid changes of alveolar type Ⅱ cell. Those changes induced by paraquat administration were mild in the rat post-treated with BHT(20mg/kg body weight) for 5 days. 3. Ultrastructural changes in the paraquat-treated rats were cytoplasmic vacuolar degeneration of the alveolar type Ⅰ cells, development of lamellar bodies and rough endoplasmic reticulum in the alveolar type Ⅱ cells, development of rough endoplasmic reticulum in fibroblasts and necrosis of the capillary endothelial cells. Those lesions induced by paraquat administration were mild in the rat post-treated with BHT(20mg/kg body weight) for 5 days.
