In order to detect the blocking factors in the sera of guinea pigs sensitized with the killed bacilli of Mycobacte rium bovis (AN5), M. avium(serotype 2 and M. tuberculosis, leukocyte adherence inhibition(LAI) test using purified protein derivatives(PPD) tuberculin as antigen was applied and a series of experiments were performed to elucidate the factors affecting the blocking effects in the LAI system. The results obtained throughout the experiments were summarized as follows: 1. It was found that blocking factors inhibiting the activity of LAI test exsisted in the sera of guinea pigs sensitized Mycobacterium spp. Blocking index was 76.0±7.4 to 54.6±7.3 in the homologous system and 8.3±0.8 to 40.8±3.9 in the heterologous system, showing the prominently high value in the homologous system. 2. When the blocking effects were examined weekly in the homologous system, the blocking index was found to be 25.8±4.1 to 49.6±5.3 at the 4th week, 71.6±5.4 to 83.2±5.6 at the 8th week and 54.3±7.6 to 62.8±5.2 at the 12th week after sensitization, appearing the highest index at the 8th week after sensitization. 3. The optimal dilution rate of serum for detecting the blocking factors in the sera of guinea pigs sensitized M. avium(serotype 2) was 1:1 to 1:3. 4. By the experiments for elucidating the mechanism of blocking activity using the leukocyte and serum from the guinea pigs sensitized with M. bovis(AN5) and homologous PPD antigen, it was evident that the blocking effects in LAI system were induced by inhibition of effector cells by blockade of PPD antigen with antibody or leukocyte with serum factors.
