Studies on the pathogenicity and effect of iron on the pathogenicity of vibrio vulnificus

The septicemia associated with Vibro vulificus has been recentyly recognized in the medical field, Therefore, these studies were proposed and carried out to determine the pathogenicity and effect of iron on pathogenicity of V. vulnificus. The results obtained were summarized as follow: 1. The LD₅₀ value of V. vulnificus injected with O.1 ml dose to 8-10 week old ICR-JCL female mice by subcutaneous route was 3.44×10⁷ in ATCC27562 strain, 2.08×10⁶ in C7184 strain, 1.35×10⁹ in KNIH-85-22 strain isolated and 4.23×10⁷ in KNIH-85-B-27 strain isolated, but by intraperitoneal route the virulence seemed to be slightly higher showing 1.09×10⁶ in ATCC 2762 strain, 6.57×10⁵ in C7184, 4.27×10⁶ in KNIH-85-22 strain isolated and 3.29×10⁶ in KNIH-85-B-27 strain isolated. The haematocrit value injected with C7184 strain, KNIH-85-22 strain isolated and KNIH-85-B-27 strain isolated to 5-6 week old female mice injected by intraperitoneal route had increased up to value of 67-68% after 3 to 6 hours from the value of 44-46% in the normal mice. The bacteremia was observed within 10 to 15 minutes in the blood stream of the mice and it was 10³ to 10⁵ cells per ml of the mice blood just before died. 2. Infection with V. vulnificus resulting in septicemia and high mortality have been correlated with pre-existing liver disease. As these conditions are associated with impaired metabolism and as iron availability in the host has been implicated in the pathogenicity of a number of bacterial infections, the role of iron as a possible factor in the pathogenesis of V. vulnificus was examined. Injection of mice with iron resulted in a lowering of the LD₅₀ from 1.09×10⁶/0.1㎖ to 2.3×10³/0.1㎖. By damaging livers with minjections of CCl₄, the LD₅₀ was 0.32×10¹/0.1㎖. The effects of adding iron on the growth of V. vulnificus in human sera was also examined. Iron appeared to be the limiting factor in the ability of this organism to survive or grow in various condition. These results, both in vito and in vivo, provided strong evidence that may play a role in the pathogenesis of V. vulnificus.
