Total vitamin A, retinol and retinyl ester in the plasma of Korean native cattle in a slaughter house at Chonju city and Chonbuk Breeding Stock Farm were determined by colorimetric method using trifluoroacetic acid and fluorescence spectrophotometry employing extraction and chromatographic separation of retinol on alumina colums. Fluorescence spectrum and the concentration of holo-retinol binding protein which reflect plasma retinol concentrations very closely were also investigated in each of the same plasma. The results obtained were summarized as follows; 1. The mean value of plasma total vitamin A concentration in Korean native cattle in summer season was 369.62(65-1260)㎍/100㎖ and retinol content remained fairly constant as 42.42±17.18㎍/100㎖ that corresponds to 141.4IU/100㎖. 2. Plasma retinyl ester concentration was 320.99㎍/100㎖ and its range was 36.50-1,201.51㎍/100㎖. 3. Fluorescence spectrum of plasma dilated to 1:100 with 0.1M NaCI showed maximum peak of fluorescence at excitation wavelength 294nm and emission wavelength 334nm. There was no any peak of fluorescence at excitation wavelength 335nm and emission wavelength 458nm reported as fluorescence peak of holo-RBP previously. 4. Concentrations of holo-RBP calculated from retinol concentrations were 32.51±12.92㎍/㎖ and values of holo-RBP by fluorometric method using excitation wavelength 335nm and emission wavelength 458,excitation wavelength 294nm and emission wavelength 334nm were 33.14±6.34㎍/㎖ and 33.93±4.20㎍/㎖, respectively. But these values by fluorometry were not found to be significantly different. Concentration of holo-RBP by fluorometric method was somewhat higher than that of holo-RBP calculated from retinol concentration.
