Studies on two-dimensional thin layer chromatographic analysis of deoxynivalenol in feed

These studies have been carried out in toxicology laboratory at Cornell University, on the determination of deoxynivalenol(DON) in the extracts of feed contaminated with molds. The extracts were prepared by the solution of methanol and water(40+60). Samples and standards were spotted on silica gel G TLC plates. Spotted plates were developed by two-dimensional TLC with toluene:ethyl acetate:formic acid(6:3:1) in the first direction and ethyl acetate:methanol:ammonium hydroxide(8.5:1:0.5) in the second direction. Separated deoxynivalenol spots from sample and standards developed in both directions were free from background interference. The quantity of deoxynivalenol present in the sample was established by visual comparison of the deoxynivalenol sizes of sample and standard DON spots. For this procedure, the standard DON plate was prepared for 50ng/g feed, 100ng/g feed, 200ng/g feed, 300ng/g feed, 400ng/g feed and 500ng/g feed. GC/MSD has been used to confirm the deoxynivalenol from the samples. Among ninety samples of feed contaminated with molds, deoxynivalenol was extracted from sixty one samples and the quantity range of the deoxynivalenol was shown from 50ng/g feed to 400ng/g feed.
