The olaquindox is used as one of the growth-promoting feed additives in pig industry. To date its maximum residue limit in pork tissues is not established. The olaquindox exists in the body both as an intact form and metabolized form such as olaquindox-4-monoxide and desoxyolaquindox. For food safety and to establish the guideline for the safe use of the olaquindox, it was demanded to develop the new analysis methods to detect the olaquindox and its metabolits in tissues. In this study, olaquindox and its metabolites were extracted with acetonitrile from muscle, liver and kidney from olaquindox-fed pigs. The extracts were then reduced to desoxyolaquindox by using ferrous sulfate. Desoxyolaquindox was treated with bis (trimethylsilyl) trifluoroacetamide (BSTFA) to obtain the derivative for GC/MS analysis. The retention time of the derivative analyzed by GC/MS was about 13.9 minutes and monitored ion masses were 303, 200 and 143. The relative ion intensity ratio was 31 : 41 : 100. The recovery rate of the olaquindox from spiked pork tissues was ranged from 82.7 to 121.1%. The study revealed that this new analysis method was more accurate compared to the conventional method. It is suggested that thisnew analysis method may replace the conventional one.
