This study was undertaken to develop the monoclonal antibodies (MAbs) for lymphocytes of Korean native cattle by the cell hybridization of myeloma NS-1 and spleen cells from Balb/c mice hyperimmunized with bovine peripheral blood T lymphocytes or thymocytes. Three MABs, KCT-2, KCT-3 and KCT-12, were developed. The isotype of these MABs was mouse Ig G2b, Ig G2b and Ig Gl, in order. Positive range of these MABs to mononuclear cells(MNC) from lymph node, spleen and thymus was 32.5~42.8%, 40.8~45.0% and 31.3~35.5%, respectively. That of these MABs to nylon wool nonadherent- and adherent-peripheral blood lymphocytes (PBL) was 46.6~56.8% and 28.2~33.7%, respectively. These MABs positive cells on frozen tissue section by immunoperoxidase stain were located in the medulla of the thymus and in the paracortical area and the mantle zone of germinal center of the lymph node. Above results indicated that these MAbs were useful tools investigate functional activities of T-lymphocyte subpopulations in the Korean native cattle.
