A survey on the presence of sulfonamides residues in raw milk has been conducted in Kyeonggi province, Korea. A total of 519 farm milk samples screened by TTC reduction test, disc assay(Bacillus stearothermophilus plate) and enzyme immunoassay respectively. Antimicrobial substances were detected from 7 samples (1.3%) by TTC test and 44 samples (8.5%) by disc assay. Sulfamethazine (SMT)-EIA Kit was used to screen sulfamethazine residues from 92 samples. Twelve screened samples were analyzed by high performance liquid chromatography (HPLC). Samples (0.5ml) were blended with octadecylsilyl (C₁₈ T) derivatized silica (2g). A column made from the sample C₁₈ T matrix was washed with hexane (8ml) and eluted with methylene chloride (8ml). The elutes were evaporated, centrifuged and detected by ultraviolet absorption at 270nm. Only one sample (No. SK-339) was contained 552ppb (ng/ml) of SMT and 2 samples were contained unknown interfering peaks. Others were not detected. Sulfonamides identity has confirmed by photo diode-array and spectral library comparison.
