The reductive metabolism of an antibacterial nitrofuran, furazolidone[N-(5-nitro-2-furfurylidene)-3-amino-2-oxazolidone] was investigated. E. coli rapidly metabolized furazolidone to the reduced cyano metabolite under aerobic condition. The major solvent-extractable metabolite produced under the aerobic incubation was isolated and analyzed by mass spectrometry. The mass spectra indicated that this metabolite was unequivocally identified as 3-(4-cyano-2-oxobutylideneamino)-2-oxazolidone, an end product of the reductive metabolism of the nitro group of furazolidone. When furazolidone was administered orally to rats and piglets, 3-(4-cyano-2-oxobutylideneamino)-2-oxazolidone was isolated and purified from the muscle, kidney and liver tissues according to the withdrawal time. These purified extracts were analyzed by high performance liquid chromatography(HPLC).
