Capsular polysaccharides(CPS) of serotypes 1, 2 & 5 of Acinobacillus pleuropneumonie were prepared from 18 hr culture supernatants by precipitaion with hexadecyltrimethyl-ammonium bromide(Cetavlon) followed by extraction with sodium chloride, reprecipitation in ethyl alcohol and phenol extraction. The CPS were evaluated as antigens for the detection of serotype-specific antibodies to A. pleuropneumoniae in pig sera by an enzyme-linked immunosorbent assay(ELISA). Serotype-specific reactions were detected in pig sera with CPS antigens of serotype 1 and 5 but serotype 2 CPS antigen was slightly cross-reactive to serotype 5 antisenrum. The herds classified as posi-tive or chronically infected to A. pleuropneumoniae were readily identified by the ELISA, with all sampled pigs showing high levels of reactivity, while the clinically negative herds were found to be ngative. In conclusion, diagnosis of pleuropneumonia caused by A. pleuropneumoniae serotypes 1, 2 & 5 can be made by screening for antibodies using CPS antigen in ELISA.
