Basolateral and brush border membrane vesicles (BLMV and BBMV) of renal proximal tubules were prepared from adult male New Zealand White rabbits to evaluate as a potential model for the assessment of ochratoxin A (OTA)-induced nephrotoxicity. PAH uptkes using BLMV, glucose and leucine uptakes using BBMV were measured in the rabbits treated with OTA. In addition, urinalysis and histopathological studies were performed to investigate the correlationship with membrane vesicle uptakes. In the uptake experiments, OTA decreased PAH uptakes in BLMV, glucose and caused cellular sweling and congestion of renal proximal and distal tubules. Taken together, results obtained by renal membrane vesicle uptake system corresponded weil with urinalysis and histopathologic findings, and suggest that this uptake system may be valuable for evaluating the nephrotoxicity of toxicants including OTA.
