The goal of this study was set to establish the molecular tools for typing of Mycobacterium bovis strains. Various M. bovis strains from different sources were typed by several typing methods including restriction fragment length polymorphism(RFLP) analysis and polymerase chain reaction(PCR). Probes for the RFLP analysis were derived from IS6110, IS1081, and direct repeat(DR) sequences. In addition, the PCR-based variable number tandem repeats(VNTR) typing was employed to differentiate M. bovis strains. Of nine M. bovis strains examined using RFLP and VNTR, seven M. bovis strains had different patterns, and two field isolates from Mexico were not distinguishable, thus indicating the same origin. The results from our study clearly show that M. bovis strains can be effectively typed by employing these molecular typing methods.
