The present study was carried out to detect DNA microsatellite for parentage verification of horses by fluorescence-based automated genotyping systems and a parentage test in Thoroughbred was conducted by the use of twelve microsatellite markers that are highly polymorphic and informative. Alleles were designated according to the number of base pairs detected in the fragments amplified by PCR. Alleles size were ranged from 148bp to 162bp fragments in AHT4, AHT5 130bp~142bp, ASB2 240bp~254bp, HMS2 218~226bp, HMS3 152bp~172bp, HMS6 159bp~169bp, HMS7 171bp~183bp, HTG4 129bp~139bp, HTG6 82bp~106bp, HTG7 120bp~128bp, HTG10 93bp~111bp and VHL20 87bp~133bp. However, no specific variation observed in Thoroughbred and Pony breeds. Among 3 foals tested for parentage verification using twelve microsatellite, Foal 1 and Foal 2 were found to be Mendelian inheritance in all loci, while Foal 3 was not marked in AHT5, HMS3, HMS6, HTG10 and VHL20.
