타우린은 동물조직 및 체액에 널리 분포하며 생체내에서 담즙배출인자 및 신경전달물질로 작용하는 세포질내이 주요한 유리 아미노산이다. 생체시료중 타우린의 정량을 위하여 기존의 방법보다 더욱 효과적인 PITC 유도체분석법을 확립하였다. 최적분석조건은 reversed phase column을 고정상으로 이용하고 sodium phosphate 와 ethyleneglycol-bis-tetraacetic acid 및 acetonitrile의 혼액을 이동상으로 하여 UV 254nm 에서 분석하였을 때 우수한 분리능을 얻을 수 있었다. PITC 유도체법에 의한 타우린의 농도별 회수율은 99.6~101.2%, 변이계수는 0.9~1.2%로 나타났으며, 시판 계란중에서 측정한 타우린 함량은 37.1±1.2nmol/g이었다.
Taurine is one of the most abundant and ubiquitous free intracellular amino acid in the fluids and various tissues of animals, and much evidences suggest that it play neurotransmitter or conjugator of bile. In this study we conducted to quantify the taurine in complex biolocal materials such as egg using high performance liquid chromatograph. Reversed phase column was chosen and combined with 10mM sodium phosphate, acetonitrile and 2mM ethyleneglycol-bis-tetraacetic acid as a mobile phase. The taurine was pre-column derivatized with phenylisothiocyanate reagent and measured absorbance at 254nm on UV detector. Recovery rates of derivatization method of the taurine from spiked egg were ranged 99.6 to 101.2% according to the concentrations. The analysis in egg revealed a coefficient of variation for reproducibility 0.9~1.2%. The levels of taurine in egg, as determined by present method, were 37.1±1.2nmol/g.
