Sandwich ELISA에 의한 vibrio vulnificus cytolysin의 검출

Detection of vibrio vulnificus cytolysin by sandwich ELISA

V. vulnificus cytolysin을 검출 하기 위한 Ezyme-Linked Immunosorbent Assay(ELI-SA)에 대한 연구에서 다음과 같은 결과를 얻었다. V. vulnificus 균 배양시 4시간에 cytolysin 활성이 최고치에 이른 후 급격히 소실되었다. 따라서 균 배양 4시간째 배양 상청액으로부터 ammonium sulfate 침전, DEAE cellulose chromatography 및 phenyl-Sepharose chromatography을 통하여 cytolysin를 순수하게 정제하였으며, 정제된 cytolysin 단백으로 닭과 토끼를 이용하여 cytolysin에 대한 monospecific polyclonal 항체를 조제하였다. Cytolysin에 대한 polyclonal 항체들은 immunoblotting 상에서 균 배양 상청액 단백들중 단지 cytolysin만 결합함으로 높은 항원 특이성을 갖고 있음을 알 수 있었다. 따라서 cytolysin에 대한 polyclonal 항체들은 cytolysin-Affi-Prep immunoaffinity chromatography을 실시하여 더욱더 정제됨으로써 sandwich ELISA의 capture 또는 detector 항체로 충분히 이용될 수 있었다. 본 연구에서 확립한 sandwich ELISA 법에서 V. vulnificus cytolysin의 검출 한계는 10 ng 이었고, 50 ng에서 포화에 도달하였다.

The sensitive Enzyme-Linked Immunosorbent Assay (ELISA) for the detection of vibrio vulnificus cytolysin was established. Cytolytic activity in V. vulnificus culture supernatant reached to the maximum about 4 hours and decreased rapidly afterwards. V. vulnificus cytolysin was purified from culture supernatants by ammonium sulfate precipitation, DEAE-cellulose chromatography and phenyl-Sepharose chromatography. The polyclonal antibodies against the purified cytolysin were produced from chicken and rabbit. Resulting antibodies showed a high antigen-specificity for cytolysin, visualing a band of 51 kDa from the V. vulnificus culture supernatants on immunoblottings. Anti-cytolysin polyclonal rabbit and chicken antibodies were used as a capture or a detector antibody in sandwich ELISA after further purification by immunoaffinity chromatography. The limit of detection was 10 ng of V. vulnificus cytolysin, with saturation achieved at 50 ng.
