랫드 조직과 혈중 항산화효소인 catalase, glutathione (GPX) 및 superoxide dismutase (SOD)의 활성을 조사하고, 멜라토닌을 투여하여 24시간 후 항산화효소의 활성에 미치는 영향을 조사하였다. 정상 조직 중 catalase 활성은 간장, 신장, 혈액에서 높은 활성을 나타내었고, 심장, 폐, 그리고 뇌는 활성이 매우 낮았다. 정상 조직중 GPX활성은 신장, 간장, 그리고 폐에서 높은 활성을 나타내었으며, 심장, 뇌에서는 활성이 낮았다. 정상 조직중 SOD활성은 간장에서 높은 활성을 나타내었으나, 신장, 혈액, 뇌, 폐, 그리고 심장에서는 활성이 비교적 낮았다. 멜라토닌의 투여로 신장과 혈액의 catalase활성이 증가하였으며 (P<0.01), 간장과 심장의 catalase활성은 높은 농도의 멜라토닌 투여로 감소하였다 (P<0.05, 0.01). 멜라토닌의 투여는 GPX활성을 신장, 폐, 간장, 뇌, 그리고 혈액에서 증가시켰다 (P<0.05, 0.01). 멜라토닌의 투여는 SOD활성을 신장과 폐에서는 증가시켰으며, 간장과 심장에서는 활성이 감소하였다 (P<0.05, 0.01).
We investigated the activities of antioxidant enzymes, such as catalase, glutathione peroxidase (GPX), and superoxide dismutase (SOD) in the tissues and the blood in rats, and the effects of melatonin on their activities in vivo by spectrophotometry or fluorophotometry. The total number of 15, four month olds male rats were divided into three groups. The animals of the treated groups were intraperitoneally injected 5 or 10 mg/kg body weight of melatonin. The animals were sacrified at the 24 hours after the treatment. The enzyme activity of catalase in cintrol rats was shown very high in the liver, kidney, and the blood, but its activity was shown relatively low in the heart, the lungs, and the brain. The GPX activity was observed high in the kidney, the liver, and the lungs, but its activity was low in the heart, the brain, and the blood. The SOD activity was observed relatively high in the liver, but its activity was low in the kidney, the blood, the brain, the lungs, and the heart. The catalase activity in the kidney and the blood in the melatonin-treated group was increased, but its activity was decreased in the liver and the heart of the high dosed group. The GPX activity in the kidney, the lungs, the brain, the liver, and the blood in the melatonin-treated group was increased in a dose-dependent manner. The SOD activity in the kidney and the lungs in the melatonin-treated group was increased, but its activity was decreased in the liver and the heart in the melatonin-treated group. These results suggested that melatonin can modulate the several antioxidant enzymes activities in rats.
