KB(kenacid blue) 시험을 이용한 광독성(phototoxicity) 대체 in vitro 시험 평가 연구

Kenacid blue assay as an in vitro alternative method for phototoxicity evaluation using human fibroblast

This study was conducted to assess a prospective altemative method as replacements for in vivo phototoxicity test. The human fibroblasts were exposed to several phototoxic chemicals(promethazine, neutral red, chlortetracycline, amiodarone, bithionol, 8-methoxypsoralen) and a non-phototoxic substance, ammonium laureth sulfate and irradiated with 5J/㎠ of UVA. The cell viability was measured by KB(kenacid blue)assay. The phototoxic potential of test chemicals in the KB PT(phototoxicity test) was assessed by determining the PIF(photoirritancy Factor) by using a cut-off value of 5. The KB PT responses of most chemicals showed a close agreement with in vivo response except chlorotetracycline. There was a relatively good agreement between in vitro KB assay and in vivo data. These results suggest that KB assay using fibroblast could be used to predict the phototoxicity of the chemicals.
