랫드에서 L- lysine 에 의한 insulin-like growth factor(IGF)-I 농도, IGF binding proteins 특성 및 IGFs carrier proteins에 미치는 효과

필수 아미노산의 일종인 L-lysine투여시 생체내 IGF-I, IGFBPs의 특성 및 carrier proteins의 변화를 관찰하기 위하여 본 연구를 시행한 바 다음과 같은 결론을 얻었다. 랫드에서 단기간 L-lysine 투여 후 100분에 혈액내 total과 free IGF-1 농도가 최고치로 증가되었고, 간장에서는 투여 후 30분에 total IGF-1 농도가 최고치로 증가되었다. 랫드에서 장기간 L-lysine을 투여한 실험군은 대조군에 비해 증체율이 유의하게 증가되었다(p<0.05). L-lysine 투여군의 혈액내 total IGF-I 농도는 대조군에 비해 증가하였으나 free IGF-I 의 농도는 억제되었다(p<0.05). L-lysine투여군의 간장과 신장에서 total IGF-I 농도, IGFBP-3와 IGFBP-2는 대조군에 비하여 증가하였고 간장에서 IGFBP-3 역시 증가하였다. 그러나 신장에서 L-lysine 투여군이 대조군에 비해 IGFBP-4가 억제되었다. L-lysine 투여군의 혈액내 150kDa carrier proteins는 대조군에 비하여 증가하였으며 50 kDa carrier proteins는 변화가 없었다. 따라서 랫드에 L-lysine의 투여는 생체내 IGF-I/IGFBPs계의 변동과 연관성이 있음을 관찰할 수 있었다.
The purpose of this study was to examine the short and long term effects of L-lysine on concentration of insulin-like growth factor-I (IGF-I), characteristics of insulin-like growth factor binding proteins (IGFBPs) and IGFs carrier protein in rat. In short-term, total and free IGF-I concentrations in serum were the highest at 100 minutes, and total IGF-I concentration in liver was the highest at 30 minutes after subcutaneous injection L-lysine (4 mg/kg body weight) in rat. In long-term, L-lysine (4 mg/kg body weight) was injected every other day for 2 weeks, body weight gain on L-lysine treated group (LTG) was increased compared with that of control group (CG) (p<0.05). Total IGF-I concentration on LTG in serum, liver and kidney was increased compared with that of CG (p<0.05). However, free IGF-I concentration in serum was decreased.In serum, amount of IGFBP-3 and -2 on LTG were increased compared with that of CG. Amount Of IGFBP-3 on LTG in liver was increased compared with that of CG 3nd IGFBP-4 in kidney was decreased. In serum, IGF-I 150 kDa carrier proteins on LTG was increased compared with that of CG. However, 50 kDa carrier proteins was not different. These results suggest that L-lysine may be effect to the IGF/IGFBPs system in rat.
