Analysis on the enterotoxigenic escherichia coli of isolated from diarrheal piglet by multilocus enzyme electrophoresis

Escherichia coli (E.coli) infection in piglets is one of the most important causes of disease-associated loss in swine production. Multilocus enzyme electrophoresis was used to assess genetic relationships and diversities amongst E. coli isolated from piglets showed the sign of diarrhea. Fifty-nine of the E. coli isolates from piglets in Chonnam, Chonbuk area were cultured in tryptic soy broth. Harvested cells were ultrasonicated to obtain the enzyme solution used as samples. These samples were runned through horizontal starch gel electrophoresis for 15~17 hours at 4℃ with 12% starch gel and 5 buffer systems. Enzyme were stained as appropriate for verification of enzyme activity. Using 17 enzyme systems, isolates were divided into 37 electrophoretic types, comprising seven major clusters, genetically distinct clusters in genetic distance 0.2. Each clusters had various serotypes. Fimbriae antigenic isolates were widely distributed across the 6 genetic clusters (a, b, d, e, f and g) and heat-stable enterotoxigenic isolates in 4 clusters (d, e, f and g). Heat-stable enterotoxigenic isolates that were in 4 clusters were of the same with 6 clusters of fimbriae antigenic isolates.
