Effect of Extract from Fermented Canavalia Gladiata on Atopic Dermatitis in an In Vitro Model

Although Canavalia gladiata has anti-inflammatory properties, the use of Canavalia gladiata is limited because it contains antinutritional and allergy causing proteins. Thus, we fermented Canavalia gladiate and investigated the effects of fermented Canavalia gladiata extract (FCE) in atopic dermatitis (AD) using in vitro model. HaCaT cells were each treated with 200, 500, 700 μg/mL of FCE, immediately followed by stimulation with 20 ng/mL tumor necrosis factor (TNF)-α, and 20 ng/mL interferon (IFN)-γ for inflammation development. We found that treatments with FCE decreased the mRNAs expression of chemokines and proinflammatory cytokines in HaCaT cells. Moreover, FCE treatment caused the suppression of Th2 cytokines and IgE secretion from AD-induced primary splenocytes. We showed that FCE treatment resulted in a decrease in histamine secretion in the PMA and A23187-simulated MC/9 cells. According to our present results, we suggest that fermented Canavalia gladiata may be effective for the treatment of AD by the attenuation of allergic reactions.
