환경오염원인 크롬의 세포독성에 대한 청미래덩굴추출물의 보호효과

Protective Effect of Smilax china L. Extract on the Cytotoxicity Induced by Chromium of Environmental Pollutant

The purpose of this study is to evaluate the cytotoxicity of chromium and protective effect of Smilax china L.(SC) extract on glucose oxidase(GO)-induced cytotoxicity in the cultured C6 glioma cells. For this study, C6 glioma cells were treated in the media containing 15~30uM concentrations of chromium trioxide(CrO₃) for 72 hours, and cell viability and lactate dehydrogenase(LDH) activity with 1,1-diphenyl-2-picrylhydrazyl(DPPH)-radical scavenging activity were assessed on these cultures. In this study, CrO₃ decreased cell viability according to the treated dosage compared with control, and than the XTT₅₀ value was determined at 30uM CrO₃ in these cultures. In the protective effect of SC extract on CrO₃-induced cytotoxicity, both of an antioxidant, catalase and SC extract significantly increased cell viability which was decreased by CrO₃-induced cytotoxicity. SC extract also showed the antioxidative effect such as DPPH-radical scaveng activity and the decreased LDH activity. From these results, it is suggested that the cytotoxicity of chromium like CrO₃ is correlated with oxidative stress of reactive oxygen species(ROS), and also SC belong to Liliaceae showed the protective effect on the chromium-induced cytotoxicity by antioxidative effect in these cultures.
