접촉성 피부염 유발제인 크롬의 세포독성 및 금은화 추출물의 영향

Cytotoxicity and Effect of Lonicerae Flos Extract against Chromium, Contact Dermatitis-Induced Agent in Cultured Human Skin Fibroblasts

To evaluate the cytotoxicity of contact dermatitis (CD)-induced agent, chromium trioxide (CrO₃) and the protective effect of Lonicerae Flos (LF) extract on the CrO₃-induced cytotoxicity in cultured human skin fibroblasts (Detroit 551). Cultured Detroit 551 cells were treated with various concentrations of CrO₃ or LF extract. In this study, CrO₃ significantly decreased cell viability compared with control, and XTT50 value (midcytotoxicity value, MCV) was determined at a concentration of 29.4uM. While, CrO₃-induced cytotoxicity was effectively prevented by vitamin E, an antioxidant. In the protective effect of LF extract on CrO₃-induced cytotoxicity, LF extract prevented CrO₃-induced cytotoxicity by the significant increase of cell viability which was decreased by CrO₃-induced cytotoxicity. And also, LF extract showed antioxidative effects such as superoxide dismutase (SOD)-like activity, the inhibitory effect of lipid peroxidation (LP) and the decrease of lactate dehydrogenase (LDH) activity. From these results, it is suggested that CrO₃ showed toxic effect on cultured Detroit 551 cells, and the cytotoxicity of CrO₃ was involved in oxidative stress. And also, LF extract effectively prevented CrO₃-induced cytotoxicity by antioxidative effect. Conclusively, the natural sustance of plants such as LF extract may be a putative resources for preventing CD mediated by CD-induced agent such as heavy metal, Cr which is related with oxidative stress.
