Protective Effect of Rumex crispus L. Extract on Cultured NIH3T3 Fibroblasts Damaged by Cadmium of Environmental Pollutant

To evaluate the protective effect of Rumex crispus L. extract on the cytotoxicity and melanogenesis of cadmium chloride (CdCl₂), environmental pollutant, cell viability was measured by 2,3-bis-[2-methoxy-4-nitro-5- sulfophenyl]-2H-tetrazolium-5-caboxanilide, disodium salt (XTT) assay after NIH3T3 fibroblasts were cultured in media containing various concentrations of CdCl₂. And also, the effect of antioxidant, vitamin E on CdCl₂-induced cytotoxicity was accessed. For the protective effect of Rumex crispus L. extract on CdCl₂-induced cytotoxicity, NIH3T3 fibroblasts were pretreated with 110 or 150μg/㎖ of Rumex crispus L. extract for 2h before the treatment of CdCl₂. And also, the antioxidative effect and melanogenesis of Rumex crispus L. extract against CdCl₂-induced cytotoxicity were assessed by 1,1-diphenyl-2-picrylhydrazyl (DPPH)-radical scavenging activity and total amount of melanin and tyrosinase activity. In this study, CdCl₂ significantly decreased cell viability in dose-dependent manner compared with control, and the XTT₅₀ value was calculated at 45.7μM of CdCl₂. In the effect of vitamin E of antioxidant, it effectively prevented CdCl₂-induced cytotoxicity by the significant increase of cell viability. In the protective effect of Rumex crispus L. extract on CdCl₂-induced cytotoxicity, Rumex crispus L. extract significantly increased cell viability which was decreased by CdCl₂-induced cytotoxicity, and also it showed the antioxidative effect by the DPPH-radical scavenging activity. In the melanogenesis, Rumex crispus L. extract showed the decreased of total amount of melanin and tyrosinase activity. From these results, it is suggested that the cytotoxicity of CdCl₂ was involved in oxidative stress, and Rumex crispus L. extract effectively prevented the cytotoxicity and the melanogenesis induced by CdCl₂. Conclusively, Rumex crispus L. extract may be a putative resources for the prevention of the toxicity and the melanogenesis of environmental pollutant such as cadmium related with oxidative stress.
