RFLP Marker Linked to Red Pericarp Color Gene of Korean Local Rice Variety, Jakwangdo

In order to identify and tag the genes for red pericarp color of rice grain, three pairs of near-isogenic lines (NILs) for the target trait were developed from the crosses between the Korean local variety with red pericarp, Jakwangdo, and three improved cultivars with white pericarp. Segregating ratio of pericarp colors in the F2 from three crosses between each pair of NILs was 3 red : 1 white. This indicated that the pericarp color genes in the parental NILs differed in one allele among two dominant genes conferring the red pericarp of Jakwangdo. To determine the genetic locus of the alleles responsible for different pericarp color in the parental NILs, RFLP analysis was carried out in the F2 populations. Twenty-one markers linked to the loci presumably responsible for pericarp color were surveyed, and none of the markers on chromosomes 1 and 6 were polymorphic but RG678 and RG477 on chromosome 7 were polymorphic between the red and the white pericarp plants. The marker RG477 among them was linked to the chromogen gene Rc on chromosome 7 with a distance of 3.2cM. Based on the results, we conclude that the genetic constitutions of these NILs would be RcRcRdRd in red and rcrcRdRd in white pericarp color.
