Agrobacterium-mediated genetic transformation of Brassica juncea cultivars has been extensively performed for the purpose of molecular breeding. B. juncea L. Czern var. Laciniata Makino, a vegetable crop also called kyona, can be transformed using its hypocotyl explants. We demonstrated that Agrobacterium-mediated transformation via vacuum infiltration moderately worked for the seedlings of B. juncea L. Czern var. Laciniata Makino according to fluorometric GUS enzyme analysis. In comparison, transformation efficiency was greatly enhanced when 3 day old seedlings were treated with hydrogen peroxide or sodium hydrosulfite solution as a chemical abrasive prior to agroinfiltration. Genetic transformation of the seedlings was confirmed by RT-PCR.
